Does the file contain binary data? If so, use fread

Try opening the file in notepad, or some other basic text editor and look at the data.
If its garbled data, its obviously in some binary format.
If its csv formatted numbers, try csvread
If its csv formatted text and numbers use xlsread
If its some other delimited numbers, try dlmread...

You can use WFDB toolbox for Matlab. It can be downloaded from http://www.physionet.org/physiotools/matlab/
