A few options depending on your situation:
1. Get the Simulink Tester toolbox. It can work with Excel spreadsheet directly.
2. Pre-process your Excel file. Write a script to import your Excel data into MATLAB workspace or save it to a .mat file. The primary function is xlsread().
3. If it is one-time, you can run uiimport to import the Excel data into MATLAB workspace.
4. You can record your interaction using uiimport in a script. This can help you develop the script.

Follow the dialog or look at the help of the "From Spreadsheet" block. It should be straightforward.
