I have a set of data in column form (size: 300x1) (class:double), is that mean my data is in array form? and how do I convert it into column vector? and how is a vector data looks like in matlab? Sorry I am new to matlab and I hope somebody can guide me in that. Thanks in advance.

Yes. It is already a column vector. There is nothing further for you to do if you want it as a column vector.
