I am doing a project in medical image processing. i have to compare automatic segmentation and manual segmentation. i have to manually segment a tumor from the brain using freehand drawing, i cannot crop it as cropping will be only in rectangular and tumor can be of any shape.i have to find the area and perimeter of tumor after doing the manual segmentation. please guide me how to segment the tumor manually using freehand drawing?

Sorry - not sure how I missed your question the first time around. Anyway, try this demo. It will let you draw a region using imfreehand(), and calculate the area and perimeter. Also does masking.
% Demo to have the user freehand draw an irregular shape over
% a gray scale image, have it extract only that part to a new image,
% and to calculate the mean intensity value of the image within that shape.
% Also calculates the perimeter, centroid, and center of mass (weighted centroid).
% Change the current folder to the folder of this m-file.
clc; % Clear command window.
clear; % Delete all variables.
close all; % Close all figure windows except those created by imtool.
imtool close all; % Close all figure windows created by imtool.
workspace; % Make sure the workspace panel is showing.
fontSize = 16;
% Read in a standard MATLAB gray scale demo image.
folder = fullfile(matlabroot, '\toolbox\images\imdemos');
baseFileName = 'cameraman.tif';
% Get the full filename, with path prepended.
fullFileName = fullfile(folder, baseFileName);
% Check if file exists.
if ~exist(fullFileName, 'file')
% File doesn't exist -- didn't find it there. Check the search path for it.
fullFileName = baseFileName; % No path this time.
if ~exist(fullFileName, 'file')
% Still didn't find it. Alert user.
errorMessage = sprintf('Error: %s does not exist in the search path folders.', fullFileName);
uiwait(warndlg(errorMessage));
return;

Hi Manas,
did you found an answer. There is a tool called bamboo tablet, that I use. However it is costly and other software based imaging tools might not give you a mask. Have you figured out a way??

imfreehand only supports gray image. for rgb image how to do it?

Why do you say that? It's not true. imfreehand() does not care what kind of image is beneath it.
